Portal vein thrombosis and arterioportal shunts: effects on tumor response after chemoembolization of hepatocellular carcinoma

- AIM: To evaluate the effect of portal vein thrombosis and arterioportal shunts on local tumor response in advanced cases of unresectable hepatocellular carcinoma treated by transarterial chemoembolization.
METHODS: A retrospective study included 39 patients (mean age: 66.4 years, range: 45-79 years, SD: 7) with unresectable hepatocellular carcinoma (HCC) who were treated with repetitive transarterial chemoembolization (TACE) in the period between March 2006 and October 2009. The effect of portal vein thrombosis (PVT) (in 19 out of 39 patients), the presence of arterioportal shunt (APS) (in 7 out of 39), the underlying liver pathology, Child-Pugh score, initial tumor volume, number of tumors and tumor margin definition on imaging were correlated with the local tumor response after TACE. The initial and end therapy local tumor responses were evaluated according to the response evaluation criteria in solid tumors (RECIST) and magnetic resonance imaging volumetric measurements.
RESULTS: The treatment protocols were well tolerated by all patients with no major complications. Local tumor response for all patients according to RECIST criteria were partial response in one patient (2.6%), stable disease in 34 patients (87.1%), and progressive disease in 4 patients (10.2%). The MR volumetric measurements showed that the PVT, APS, underlying liver pathology and tumor margin definition were statistically significant prognostic factors for the local tumor response (P = 0.018, P = 0.008, P = 0.034 and P = 0.001, respectively). The overall 6-, 12- and 18-mo survival rates from the initial TACE were 79.5%, 37.5% and 21%, respectively.
CONCLUSION: TACE may be exploited safely for palliative tumor control in patients with advanced unresectable HCC; however, tumor response is significantly affected by the presence or absence of PVT and APS.
Impact of deep convection in the tropical tropopause layer in West Africa: in-situ observations and mesoscale modelling

- We present the analysis of the impact of convection on the composition of the tropical tropopause layer region (TTL) in West-Africa during the AMMA-SCOUT campaign. Geophysica M55 aircraft observations of water vapor, ozone, aerosol and CO2 during August 2006 show perturbed values at altitudes ranging from 14 km to 17 km (above the main convective outflow) and satellite data indicates that air detrainment is likely to have originated from convective cloud east of the flights. Simulations of the BOLAM mesoscale model, nudged with infrared radiance temperatures, are used to estimate the convective impact in the upper troposphere and to assess the fraction of air processed by convection. The analysis shows that BOLAM correctly reproduces the location and the vertical structure of convective outflow. Model-aided analysis indicates that convection can influence the composition of the upper troposphere above the level of main outflow for an event of deep convection close to the observation site. Model analysis also shows that deep convection occurring in the entire Sahelian transect (up to 2000 km E of the measurement area) has a non negligible role in determining TTL composition.
Bivariate and Multivariate NeuroXidence: A Robust and Reliable Method to Detect Modulations of Spike–Spike Synchronization Across Experimental Conditions

- Synchronous neuronal firing has been proposed as a potential neuronal code. To determine whether synchronous firing is really involved in different forms of information processing, one needs to directly compare the amount of synchronous firing due to various factors, such as different experimental or behavioral conditions. In order to address this issue, we present an extended version of the previously published method, NeuroXidence. The improved method incorporates bi- and multivariate testing to determine whether different factors result in synchronous firing occurring above the chance level. We demonstrate through the use of simulated data sets that bi- and multivariate NeuroXidence reliably and robustly detects joint-spike-events across different factors.
Restriction of trophic factors and nutrients induces PARKIN expression

- Parkinson's disease (PD) is the most frequent neurodegenerative movement disorder and manifests at old age. While many details of its pathogenesis remain to be elucidated, in particular the protein and mitochondrial quality control during stress responses have been implicated in monogenic PD variants. Especially the mitochondrial kinase PINK1 and the ubiquitin ligase PARKIN are known to cooperate in autophagy after mitochondrial damage. As autophagy is also induced by loss of trophic signaling and PINK1 gene expression is modulated after deprivation of cytokines, we analyzed to what extent trophic signals and starvation stress regulate PINK1 and PARKIN expression. Time course experiments with serum deprivation and nutrient starvation of human SH-SY5Y neuroblastoma cells and primary mouse neurons demonstrated phasic induction of PINK1 transcript up to twofold and PARKIN transcript levels up to sixfold. The corresponding threefold starvation induction of PARKIN protein was limited by its translocation to lysosomes. Analysis of primary mouse cells from PINK1-knockout mice indicated that PARKIN induction and lysosomal translocation occurred independent of PINK1. Suppression of the PI3K-Akt-mTOR signaling by pharmacological agents modulated PARKIN expression accordingly. In conclusion, this expression survey demonstrates that PARKIN and PINK1 are coregulated during starvation and suggest a role of both PD genes in response to trophic signals and starvation stress.
Ion beam analysis based on cellular nonlinear networks
- The development of a non- destructive measurement method for ion beam parameters has been treated in various projects. Although results are promising, the high complexity of beam dynamics has made it impossible to implement a real time process control up to now. In this paper we will propose analysing methods based on the dynamics of Cellular Nonlinear Networks (CNN) that can be implemented on pixel parallel CNN based architectures and yield satisfying results even at low resolutions.
Extraction of network topology from multi-electrode recordings: is there a small-world effect?
- The simultaneous recording of the activity of many neurons poses challenges for multivariate data analysis. Here, we propose a general scheme of reconstruction of the functional network from spike train recordings. Effective, causal interactions are estimated by fitting generalized linear models on the neural responses, incorporating effects of the neurons’ self-history, of input from other neurons in the recorded network and of modulation by an external stimulus. The coupling terms arising from synaptic input can be transformed by thresholding into a binary connectivity matrix which is directed. Each link between two neurons represents a causal influence from one neuron to the other, given the observation of all other neurons from the population. The resulting graph is analyzed with respect to small-world and scale-free properties using quantitative measures for directed networks. Such graph-theoretic analyses have been performed on many complex dynamic networks, including the connectivity structure between different brain areas. Only few studies have attempted to look at the structure of cortical neural networks on the level of individual neurons. Here, using multi-electrode recordings from the visual system of the awake monkey, we find that cortical networks lack scale-free behavior, but show a small, but significant small-world structure. Assuming a simple distance-dependent probabilistic wiring between neurons, we find that this connectivity structure can account for all of the networks’ observed small-world-ness. Moreover, for multi-electrode recordings the sampling of neurons is not uniform across the population. We show that the small-world-ness obtained by such a localized sub-sampling overestimates the strength of the true small-world structure of the network. This bias is likely to be present in all previous experiments based on multi-electrode recordings.
Soil moisture initialization effects in the Indian monsoon system
- Towards the goal to understand the role of land-surface processes over the Indian sub-continent, a series of soil-moisture sensitivity simulations have been performed using a non-hydrostatic regional climate model COSMO-CLM. The experiments were driven by the lateral boundary conditions provided by the ERA-Interim (ECMWF) reanalysis. The simulation results show that the pre-monsoonal soil moisture has a significant influence on the monsoonal precipitation. Both, positive and negative soil-moisture precipitation (S-P) feedback processes are of importance. The negative S-P feedback process is especially influential in the western and the northern parts of India.
Regional climate projections in two alpine river basins: Upper Danube and Upper Brahmaputra
- Projections from coarse-grid global circulation models are not suitable for regional estimates of water balance or trends of extreme precipitation and temperature, especially not in complex terrain. Thus, downscaling of global to regionally resolved projections is necessary to provide input to integrated water resources management approaches for river basins like the Upper Danube River Basin (UDRB) and the Upper Brahmaputra River Basin (UBRB).
This paper discusses the application of the regional climate model COSMO-CLM as a dynamical downscaling tool. To provide accurate data the COSMO-CLM model output was post-processed by statistical means. This downscaling chain performs well in the baseline period 1971 to 2000. However, COSMO-CLM performs better in the UDRB than in the UBRB because of a longer application experience and a less complex climate in Europe.
Different climate change scenarios were downscaled for the time period 1960–2100. The projections show an increase of temperature in both basins and for all seasons. The values are generally higher in the UBRB with the highest values occurring in the region of the Tibetan Plateau. Annual precipitation shows no substantial change. However, seasonal amounts show clear trends, for instance an increasing amount of spring precipitation in the UDRB. Again, the largest trends for different precipitation statistics are projected in the region of the Tibetan Plateau. Here, the projections show up to 50% longer dry periods in the months June to September with a simultaneous increase of about 10% for the maximum amount of precipitation on five consecutive days. For the Assam region in India, the projections also show an increase of 25% in the number of consecutive dry days during the monsoon season leading to prolonged monsoon breaks.
